If you activate Conquistador of the golden land as CL1, and Twin Twisters is chained as CL2, destroying Conquistador, Conquistador won't summon itself and you won't be able to destroy a card with it. A continuous trap card must be on the field at resolution to resolve with an effect.

First of all Mystic Mine doesn't negate effects, it prevents activations. When you activate these Golden Land trap cards, all the effect resolves as the effect of the trap card you activate, which is summoning and destroying/banishing. This second part is not a monster effect.

why do you even care

The card's text isn't updated with PSCT, since its printing is very old. The first one is a trigger effect which activates when the monster is normal summoned, starting a chain.
